THE N-TERMINAL ARMS OF TWO MONOMERS ARE INTERTWINED IN FORMING THE CENTRAL PAIR OF STRANDS IN A 12-STRANDED SHEET OF THE DIMER. PROTEOLYTIC PROCESSING CLEAVES THIS PAIR OF STRANDS TO RELEASE THE ACTIVE TOXIN AS A MONOMER, WHICH EXTENDS FROM THE NPS SITE TO THE CPS SITE WHEN PROTEINASE K IS USED FOR THE PROCESSING.
